Strain Description

The ALB gene encodes the most abundant protein in human blood, which functions to regulate plasma colloid osmotic pressure and as a carrier protein for a variety of endogenous molecules, including hormones, fatty acids, and metabolites, as well as exogenous drugs. In addition, the protein exhibits esterase-like activity and has broad substrate specificity. The peptide EPI-X4, derived from this protein, is an endogenous inhibitor of the CXCR4 chemokine receptor.

This strain carries a mouse Alb gene promoter-driven improved Cre (iCre) expression element insertion. When H11-Alb-iCre mice are crossed with mice containing loxP sites, Cre recombinase-mediated loxP site sequence recombination is induced in liver tissue. The heterozygous H11-Alb-iCre mice are viable and fertile.

 

The Alb Promoter-iCre gene expression element was inserted into the H11 Safe Harbor Locus in C57BL/6J mice.

Detection of Cre expression

a. Method

H11-Alb-iCre mice were mated with Rosa26-LSL-tdTomato mice to produce double heterozygous offspring mice, which are expected to express red fluorescence in the Cre-positive cells. Liver and stomach tissues were collected, frozen sectioned, and then observed for fluorescence onset.

b. Genotype

Experimental group (upper): H11-Alb-iCre[KI/+];Rosa26-LSL-tdTomato[CKI/+]
Control group (bottom): Rosa26-LSL-tdTomato[CKI/+]

c. Result

1. Liver

2. Stomach

Description: Intense tdTomato orange fluorescence was detected in a large number of hepatocytes and partially in the stomach.

Conclusion: This strain shows intense recombination in hepatocytes and partial fluorescence in gastric tissues (data shown). Also, recombination bands are visible in other tissues (lung, kidney, intestine, adipose tissue) (data not shown).

The transgenic element of this strain is located on chromosome 11. Please avoid using flox mice with the transgenic locus on chromosome 11 for breeding.
